No... the final result would be very different if a brush is based on only 4 squares (since the entire stroke is stretched around another shape). You are surely thinking of the Artistic Media Sprayer tool, where you can make one object and then spray it along an existing curve of any length. But again, the result is different yet again with the Sprayer: the corners would not conform the way they do in the video.
